MOLECULAR CHARACTERIZATION OF Aspergillus flavus TOXIGENICITY IN AGRICULTURAL COMMODITIES IN INDONESIA

چکیده
Toxigenic Aspergillus flavus is a primary producer of aflatoxin in Indonesia, and its presence can lead to the contamination agricultural commodities. This poses risk export-targeted commodities, potentially resulting their rejection. Therefore, this study aims characterize molecular profile nativeA. isolated from several Indonesian products, with major focus on toxigenicity toxin production. A total 18 A. collections were nutmeg, ground peanut, cacao, coffee bean, corn, white pepper, soil peanut plantation. Species identification was carried out using morphological approaches. The isolates characterized based amplification gene clusters, while production assessed through growth simulation 10% coconut broth media followed by HPLC quantification. result showed that all confirmed as sequence analysis ITS region. 11 (61%) toxigenic produced 1-2 types aflatoxin, varying concentrations high, moderate, or low levels AFB1. High AFB1 seven namely BIO3313, BIO33212, BIO3361, BIO33404, BIO3338, BIO3352, BIO3344, had concentration ranging 76.78 2241.06 µg/kg, three (BIO3314, BIO3312, BIO3381) below 1 µg/kg. Twenty-nine pairs gene-specific sequences successfully amplified single band, some non-specific patterns non-toxigenic isolates. Based results, it concluded completed clusters variations deletion observed both However, no specific target could effectively distinguish two groups. Two BIO3393 BIO33403 exhibited large be potential candidates for biocontrol agents.

Characterization of aflatoxigenic Aspergillus flavus and A. parasiticus strain isolates from animal feedstuffs in northeastern Iran
Aflatoxins are secondary toxic metabolites produced by some Aspergillus spp. particularly, Aspergillus flavus and A. parasiticus that contaminate food and feed. The objective of this study was to evaluate the contamination of feedstuffs with Aspergillus spp. and detect genes involved in the aflatoxin biosynthesis pathway of A. flavus and A. parasiticus isolates. A total of 110 cow feed samples ...
متن کاملDetection of Aflr Gene and Toxigenicity of Aspergillus flavus Group Isolated from Patients with Fungal Sinusitis
Background: Aspergillus flavus is the second most important Aspergillus species causing human infections particularly fungal sinusitis. Since little is known about aflatoxin producing ability of clinical isolates, this study was undertaken to detect the aflatoxigenic isolates amongst these isolates. Methods: A total of 23 isolates of A. spp. which were recovered from patients proved to have fun...

Fifteen non-aflatoxigenic strains of Aspergillus flavus, represent a wide range of geographic regions of Iran (six provinces include Fars, Ardebil, Guilan, Golestan, Kerman and Semnan) and vegetative compatibility groups (VCGs), were collected from corn (Zea mays L.), peanut (Arachis hypogaea L.) and pistachio (Pistachia vera L.) soils and kernels, were screened for the presence of aflatoxin bi...
متن کاملCharacterization of the Far Transcription Factor Family in Aspergillus flavus
Metabolism of fatty acids is a critical requirement for the pathogenesis of oil seed pathogens including the fungus Aspergillus flavus Previous studies have correlated decreased ability to grow on fatty acids with reduced virulence of this fungus on host seed. Two fatty acid metabolism regulatory transcription factors, FarA and FarB, have been described in other filamentous fungi. Unexpectedly,...
